چکیده

By Pilar Heredia-Middleton Washington State University December 2005 Chair: Gary Thorgaard Several review articles have been published on the role of heat shock proteins, and their link to temperature in fish and other organisms. Heat shock protein 70 (70KDa) is the most commonly-expressed protein in response to thermal stress, and its expression has been associated with differences in thermal habitats. This thesis presents a general background and review of heat shock proteins, followed by a review of aquatic and marine organisms’ response to thermal stress. In addition, I described the heat shock response to thermal stress of Oncorhynchus mykiss rainbow trout clonal lines; developed from individuals from different habitats. The experiment characterized response of three clonal lines, Arlee, OSU and Whale Rock. The expression patterns of hsp70 RNA from blood were quantified using real-time PCR. The hsp70 RNA expression patterns of these clonal lines were found to be significantly different. The most striking result was the lack of any detectable response by the Arlee line. Both Whale Rock and OSU hsp70 expression patterns showed a maximum induction temperature at 22°C; Whale Rock

DNA fingerprinting confirms isogenicity of androgenetically derived rainbow trout lines.

Homozygous and hybrid clonal lines of rainbow trout (Oncorhynchus mykiss) were confirmed to be isogenic using multilocus DNA fingerprinting. Homozygous clones were produced by androgenesis and gynogenesis using gametes from androgenetic male and female rainbow trout, respectively. Isogenic F1 hybrid lines were produced by crossing homozygous fish from different strains. One line of hybrid clone...

Effects of heat shock and hypoxia on protein synthesis in rainbow trout (Oncorhynchus mykiss) cells.

We examined the effects of heat stress (from 18 degreesC to 26 degreesC) and low oxygen tension (1% O2=1 kPa) on protein synthesis in primary cultures of hepatocytes, gill epithelial cells and fibroblast-like RTG-2 cells of rainbow trout Oncorhynchus mykiss. All these cell types displayed elevated levels of 67, 69 and 92 kDa proteins, whereas a 104 kDa protein was induced only in RTG-2 cells. H...

Dataset of proinflammatory cytokine and cytokine receptor gene expression in rainbow trout (Oncorhynchus mykiss) measured using a novel GeXP multiplex, RT-PCR assay

A GeXP multiplex, RT-PCR assay was developed and optimized that simultaneously measures expression of a suite of immune-relevant genes in rainbow trout (Oncorhynchus mykiss), concentrating on tumor necrosis factor and interleukin-1 ligand/receptor systems and acute phase response genes. The dataset includes expression values for drpt, il11a, il1b1, il1b2, il1b3, il1r-like-1(e3-5), il1r-like-1(e...

Glucocorticoid-mediated attenuation of the hsp70 response in trout hepatocytes involves the proteasome.

The physiological implication of elevated cortisol levels on cellular heat-shock protein 70 (hsp70) response was examined using primary cultures of rainbow trout (Oncorhynchus mykiss) hepatocytes. Trout hepatocytes treated with cortisol, the predominant glucocorticoid in teleosts, responded to the heat shock (+15 degrees C for 1 h) with a significant drop in hsp70 accumulation over a 24-h recov...
